RHINOLOPHUS MEHELYI SCYTHOTAURICUS SUBSP. NOV. (RHINOLOPHIDAE, CHIROPTERA) FROM THE LOWER PLEISTOCENE OF THE TAURIDA CAVE IN CRIMEA

Abstract

A new extinct subspecies of the Mehely’s horseshoe bat, Rhinolophus mehelyi scythotauricus subsp. nov., is described on the base of an incomplete skull from the Lower Pleistocene deposits of the Taurida cave in the central Crimea. It is the largest member of the R. euryale group. In terms of the evolutionary level, it is intermediate between Plio-Pleistocene R. mehelyi birzebbugensis Storch, 1974 and recent members of the species, but its large size and relatively narrow upper molars may indicate belonging to a separate phylogenetic lineage within R. mehelyi Matschie, 1901. R. mehelyi scythotauricus subsp. nov. is the first fossil record of the species in the Crimea; it is also one of the northernmost finds of R. mehelyi.

NEW DATA ON HESPERORNITHIDS (AVES: ORNITHURAE) FROM THE CAMPANIAN OF THE LOWER VOLGA REGION (LATE CRATACEOUS, RUSSIA)

Abstract

Taxonomic diversity of the Late Cretaceous Hesperornithidae (Aves: Hesperornithidae) of the European part of Russia and Eastern Europe as a whole remain poorly understood, and the morphology of these large flightless birds is poorly known. New finds of Hesperornithidae in the Karyakino locality (Saratov oblast, Russia) confirm the coexistence of two forms of these flightless seabirds in the Campanian (mid-Late Cretaceous) of the Lower Volga region. For Hesperornis rossicus Nessov et Yarkov, 1993, a femur is described for the first time; it indicates a morphological distinctiveness of this large species as compared to the North American H. regalis Marsh, 1872.

Abstract

The development of technologies for effective targeted drug delivery for oncotherapy requires the elaboration of new methods to analyze the features of micro- and nanoscale distributions of antitumor drugs in cells and tissues. This paper presents a new approach to three-dimensional analysis of intracellular distribution of cytostatics using fluorescence scanning optical-probe nanotomography technology. Correlative analysis of nanostructure and distribution of injected doxorubicin in studied MCF-7 human breast adenocarcinoma cells made it possible to reveal the features of drug penetration and accumulation in cells. The developed technology based on the principles of scanning optical probe nanotomography is applicable to study the distribution patterns of various fluorescent or fluorescence-labelled substances in cells and tissues.

The effect of modeling septoplasty and modeling sensory deprivation of the olfactory analyzer in rats on changes in the frequency domain of heart rate variability was compared. Bulbectomy provokes more pronounced changes in heart rate variability in rats, compared with septoplasty simulation.

APPLICATION OF BIOMARKER FATTY ACIDS OF ROE AND FILLETS TO DETERMINE THE ORIGIN OF STERLET (ACIPENSER RUTHENUS, LINNAEUS, 1758)

Abstract

Significant differences were found in the fatty acid composition of roe and muscle tissue (fillet) of starlet (Acipenser ruthenus, Linnaeus, 1758) from Yenisei River and aquaculture farms associated with different food sources. Roe and muscle tissue of sterlet from the natural habitat were characterized by significantly higher levels of fatty acids – biomarkers of diatoms and bacterial matter. Sterlet grown in aquaculture conditions had in the composition of fatty acids significantly higher percentages of oleic and linoleic acids, as well as fatty acids, biomarkers of marine copepods. For the first time, it was proposed to use the calculation of the ratio of the percentage of several biomarker fatty acids and set threshold values for determining the origin of sturgeon roe and fillet from the natural habitat and aquaculture.

DISCOVERY OF SIBOGLINIDS (ANNELIDA, SIBOGLINIDAE) IN THE ESTUARIES OF THE LARGEST ARCTIC RIVERS ARE ASSOCIATED WITH PERMAFROST GAS HYDRATES

Abstract

In the estuaries of the largest Arctic rivers, namely, the Yenisei, Lena and Mackenzie, marine worms of the family Siboglinidae have been found. Their metabolism is provided by symbiotic chemoautotrophic bacteria. The strong salinity stratification characteristic of the estuaries of the largest Arctic rivers ensures high salinity at depths of 25–36 m, where siboglinids were found. High concentrations of methane necessary for the metabolism of siboglinids arise because of dissociation of permafrost gas hydrates under the influence of river runoff in the conditions of Arctic warming

FLUCTUATIONS IN THE RATE OF RADIOACTIVE DECAY OF THE NATURAL POTASSIUM-40 ISOTOPE CAN POTENTIALLY AFFECT THE PERIODIC “SPONTANEOUS” ACTIVITY OF ANIMALS

Abstract

The body temperature rhythms in laboratory mice C57Bl/6 and common greenfinches (Chloris chloris) and the moments of feeding by common starlings (Sturnus vulgaris) have been compared with the intensity of fluctuations of 40К radioactive decay. It was found that body temperature changes in greenfinches and mice positively correlate with the intensity of fluctuations of 40K radioactive decay. The method of superimposed epochs shows that the increase of body temperature in mice, reflecting the phase of the beginning of sleep/wake cycle activity, as well as food intake by starlings are observed at an increase in the intensity of 40K radioactive decay. Thus, animal activity in the ultradian range of periods may be related to external quasi-rhythmic physical influences, rather than determined only by endogenous processes. Given the extremely low dose of natural 40К exposure, a factor that can cause observed radioactivity fluctuations may be biotropic.

DISTRIBUTION OF SIBOGLINIDS (ANNELIDA, SIBOGLINIDAE) IN THE LAPTEV SEA AND ADJACENT AREAS OF THE ARCTIC BASIN

Abstract

The purpose of the article is to assess the biodiversity of gutless marine worms of the family Siboglinidae (Annelida), whose metabolism is provided by symbiotic bacteria that oxidize hydrogen sulfide and methane. Within the geographical boundaries of the Laptev Sea, 7 species of siboglinids were found, another 1 species was found in the adjacent sector of the Arctic basin. The largest number of finds and the greatest biological diversity of siboglinids were found in the eastern part of the Laptev Sea in the field of numerous methane. One find was noted in the estuary area of the Lena River at a depth of 25 m. The possible connection of siboglinids with methane seepage areas is discussed.

MULTILOCUS ANALYSIS OF PHYLOGENETIC RELATIONSHIPS IN THE CROCIDURA SUAVEOLENS SENSU LATO SPECIES COMPLEX: COMPARISON WITH MITOCHONDRIAL DATA

Abstract

For the first time, we attempt to study the phylogeny of the Crocidura suaveolens s. l. species complex using multilocus analysis. Sequencing data for 16 nuclear genes indicate the existence of several distinct forms within the species complex, and its structure does not in general contradict mitochondrial phylogeny. Siberian shrew is characterized by some specificity of the nuclear genome, but the degree of its genetic differentiation does not correspond to species-level. We clarified the relationships of Crocidura aff. suaveolens from South Gansu and Sichuan with other forms of the species complex. The shrews from Buryatia and Khentii also belong to this form, but their mtDNA had apparently introgressed in the past from C. shantungensis. In the recent time, hybridization of C. suaveolens s.str. with C. aff. suaveolens and C. güeldenstaedtii, had also occurred. Taking into account multiple introgression events in the history of C. suaveolens s.l., for the analysis of the phylogenetic relations between its forms a significant increase in the set of loci is required.

PHYLOGENY OF THE DIPUS SAGITTA SPECIES COMPLEX BASED ON THE SEQUENCING OF NUCLEAR GENES

Abstract

The northern three-toed jerboa Dipus sagitta was long considered to be a single polytypic species. Earlier, a high genetic diversity of D. sagitta was revealed on the basis of several mitochondrial and nuclear genes, and the existence of several separate species within it was hypothesized. However, a small number of available nuclear genes prevented the establishment of relationships between identified phylogenetic lineages. Here, we significantly expand the set of utilized nuclear DNA loci, which resulted in a sufficiently higher resolution of the phylogenetic tree for 10 forms of D. sagitta. The identified species structure mainly confirms the topology and relationship of the mtDNA lineages. At the same time, the mitochondrial and nuclear phylogenies are not completely consistent. Hence, a part of the genetic lineages of D. sagitta is apparently a product of reticular evolutionary processes. This taxon is thus the diverse species complex D. sagitta sensu lato, in which long-diverged lineages are not always reproductively isolated.

D2-LIKE DOPAMINE RECEPTOR DISTRIBUTION IN THE BRAIN OF RATS WITH DIFFERENT TYPES OF GENETIC EPILEPSY

Abstract

The study investigated D2-like dopamine receptor (D2DR) binding densities in cortex and striatum between rats with absence and/or audiogenic genetically determined epilepsy and normal Wistar rats by autoradiography. It was shown that D2DR binding density was lower in the dorsal and ventrolateral aspects of nucleus accumbens in epileptic vs non-epileptic rats. Additionally, audiogenic rats had higher motor, somatosensory cortex, and dorsal striatum binding as in contrast with non-audiogenic groups. This data suggests that there is a common neuronal circuit involved in the pathogenesis of generalized epilepsy of both convulsive and non-convulsive forms.

THE XMAS-2 PROTEIN OF DROSOPHILA MELANOGASTER UNDERGOES CLEAVAGE INTO TWO FRAGMENTS

Abstract

The TREX-2 complex integrates several stages of gene expression, such as transcriptional activation and mRNA export. In D. melanogaster TREX-2 consists of four main proteins: Xmas-2, ENY2, PCID2, and Sem1p. Xmas-2 protein is the core subunit of the complex with which other TREX-2 subunits interact. Xmas-2 homologues have been found in all higher eukaryotes. Previously, it was shown that the human Xmas-2 homologue, GANP protein can undergo cleavage into two parts, probably during apoptotic cell death. We showed that the Xmas-2 protein of D. melanogaster also can split into two fragments. The resulting fragments of the protein correspond to the two large domains of Xmas-2. Protein splitting is observed both in vivo and in vitro. However, Xmas-2 cleavage in D. melanogaster is observed under normal conditions and is probably a part of the mechanism of transcription and mRNA export regulation in D. melanogaster.

Abstract

A synthesis of a new series of 1-[ω-(bromophenoxy)alkyl]-uracil derivatives containing in position 3 naphthalen-1-yl-, naphthalen-2-yl-, 1-bromonaphthalen-2-ylmethyl, benzyl, and anthracene 9-methyl fragment was carried out. The antiviral properties of the synthesized compounds were studied against human cytomegalovirus. It was found that the compound, which has in its composition a bridge consisting of five methylene groups, has a high anti-cytomegalovirus activity in vitro.

STUDY OF THE IN VIVO FUNCTIONAL ROLE OF MUTATIONS IN THE BTB DOMAIN OF THE CP190 PROTEIN OF DROSOPHILA MELANOGASTER

Abstract

The Drosophila transcription factor СР190 is one of the key proteins that determine the activity of housekeeping gene promoters and insulators. CP190 has an N-terminal BTB domain that allows for dimerization. Many of known Drosophila architectural proteins interact with the hydrophobic peptide-binding groove in the BTB domain, which is supposed to be one of the mechanisms for recruiting CP190 to regulatory elements. To study the role of the BTB domain in the interaction with architectural proteins, we obtained transgenic flies expressing CP190 variants with mutations in the peptide-binding groove, which disrupts their interaction with architectural proteins. As a result of the studies, it was found that mutations in the BTB domain do not affect binding of the CP190 protein to polytene chromosomes. Thus, our studies confirm the previously obtained data that CP190 is recruited to regulatory elements by several transcription factors interacting in addition to BTB with other CP190 domains.

Abstract

The effect of different modes of feeding and lighting (natural and continuous) on the phospholipid composition of Atlantic salmon fingerlings reared under commercial aquaculture in the summer-autumn period in North Ossetia-Alania was studied. Qualitative and quantitative determination of phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol, lysophosphatidylcholine, sphingomyelin was carried out by high performance liquid chromatography. A decrease (September-November) The content of the studied phospholipids in fingerlings decreased, which should be considered primarily as a biochemical adaptation of development, preparation of juveniles for the upcoming smoltification. The effects of lighting and feeding regime on phospholipid composition were found mainly in fish reared under constant lighting and 24/7 feeding and fish reared under natural light and feeding during daylight hours, however, the observed changes were not specific to a particular experimental group of fish in the framework of this study.

EFFECT OF METHYL JASMONATE ON THE GENE EXPRESSION, ENCODING NON-PROTEIN THIOL ENZYMES IN WHEAT UNDER CADMIUM

Abstract

The effect of the phytohormone – methyl jasmonate (MJ) on the expression of the TaGS1 and TaPCS1 genes encoding glutathione synthetase and phytochelatin synthase, respectively, which are key enzymes in the synthesis of glutathione and phytochelatins in wheat (var. Moskovskaya 39), was studied. For the first time, it has been shown, that pretreatment of plants by exogenous MJ (1 μM) leads to an increase in the accumulation of transcripts of the TaGS1 and TaPCS1 genes in leaves without cadmium effect. When cadmium (CdSO4, 100 μM) was added into the nutrient solution, the transcript level of TaGS1 gene increased in plants pretreated by MJ (compared to untreated plants), while the transcript level of TaPCS1 remained unaffected. The pretreatment of plants by MJ leads to a lower accumulation of cadmium in the roots and leaves of wheat. At the same time, MJ had no effect on the linear growth parameters of plants, but had a positive effect on the accumulation of biomass under cadmium. It has been suggested that MJ is involved in plant tolerance to cadmium by increasing the expression of the TaGS1 and TaPCS1 genes and, as a result, enhancing the synthesis of chelating compounds, as well as by reducing the supply of metal ions to plants.

CHLOROPHYLL A DIMERS HARBORED BY WATER-SOLUBLE PROTEIN BoWSCP PHOTOSENSITIZE REDUCTION OF CYTOCHROME C

Abstract

When bound to water-soluble proteins of the WSCP family, chlorophyll molecules form dimers structurally similar to “special pair” of chlorophylls (bacteriochlorophylls) in photosynthetic reaction centers. Being exposed to red light (λ ≥ 650 nm) in oxygen-free solutions, chlorophyll a dimers harbored by BoWSCP holoproteins (from Brassica oleracea var. botrytis) have sensitized the reduction of cytochrome c. According to absorption and circular dichroism spectroscopy data, the photochemical process did not significantly impair the structure of chlorophyll a molecules as well as their dimers harbored by BoWSCP protein. Adding tris(hydroxymethyl)aminomethane as an electron donor for chlorophyll recovery stimulated the photoreduction of cytochrome c.

ACTIVITY OF NSP14 EXONUCLEASE FROM SARS-COV-2 TOWARDS RNAS WITH MODIFIED 3'-TERMINI

Abstract

The COVID-19 pandemic has shown the urgent need for new treatments for coronavirus infections. Nucleoside analogs have been successfully used to inhibit replication of some viruses, through the incorporation into the growing DNA or RNA chain. However, the replicative machinery of coronaviruses contains nsp14, a a non-structural protein with a 3'→5'-exonuclease activity that removes misincorporated and modified nucleotides from the 3' end of the growing RNA chain. Here we studied the efficiency of hydrolysis of RNA containing various modifications in the 3'-terminal region by SARS-CoV-2 nsp14 exonuclease and its complex with the auxiliary protein nsp10. Single-stranded RNA was a preferable substrate compared to double-stranded, consistent with the model of transfer of the substrate strand to the exonuclease active site proposed on the basis of structural analysis. Modifications of the phosphodiester bond between the penultimate and last nucleotides had the greatest effect on nsp14 activity.

INTRACELLULAR DELIVERY OF ANTIBODY-LIKE MOLECULE, CAPABLE TO INHIBIT C-MYC

Abstract

A modular nanotransporter (MNT) carrying the sequence of an antibody-like molecule, an anti-c-Myc nanobody, was synthesized and characterized. It was demonstrated that the created MNT is able to interact with the target protein, c-Myc oncogene, with a dissociation constant of 46 ± 14 nM, internalize into target cells, change Myc-dependent expression, and exert an antiproliferative effect.

COMPARATIVE DETERMINATION OF NATURAL ANTIBODIES FOR THE ASSESSMENT OF THE CARDIOVASCULAR SYSTEM AND THE LEVEL OF HUMAN

Abstract

A comparative analysis of natural antibodies to β-endorphin, angiotensin, dopamine, serotonin, parameters of the cardiovascular system and anxiety levels was carried out for 241 athletes of various qualifications and sports. The obtained indicators of the cardiovascular system were compared with reference values. A significant increase in the level of natural antibodies to angiotensin was established for all groups of athletes. In the case of dopamine, serotonin, these differences are associated with the qualification of the athlete, for β-endorphin, differences in the level of the indicator depending on the sport were found. A group of individuals with high levels of situational and personal anxiety was found among highly qualified athletes. An increase in blood pressure in athletes of cyclic sports and martial arts is adaptive, and in athletes of speed-strength sports it leads to a change in the walls of the myocardium. As a result of the study, the possibility of a comprehensive determination of natural antibodies and functional indicators as diagnostic markers for assessing the state of the human cardiovascular system has been shown.

SELECTION OF AN AMINO ACID SITE WITH ONE OF THE FASTEST CLEAVAGE KINETICS BY THE ENDOSOMAL PROTEASE CATHEPSIN B FOR POTENTIAL USE IN DRUG DELIVERY SYSTEMS

Abstract

Based on known literature data, six peptide sequences were selected that are potentially capable of being rapidly cleaved by the endosomal protease cathepsin B. For comparison, cathepsin B cleavage of common linker sequences, polyglycine and polyglycine-serine, was also studied. Different ends of these peptides were labeled with sulfoCyanine3 and sulfoCyanine5 fluorescent dyes, between which Förster resonant energy transfer (FRET) is possible. The kinetics of cleavage of peptides by cathepsin B was studied on a multimodal plate reader by FRET signal reduction. FKFL and FRRG cleavage sites have been shown to be the most suitable for potential use in various drug delivery systems. These sites are much more efficiently cleaved under slightly acidic conditions of endosomes than at neutral extracellular pH.
