Activation of Su(Hw)-Controlled Genes Is Associated with Increase in GAF Binding

Аннотация

The interaction of the GAF protein with the promoters of neuron-specific genes during activation and repression of transcription was studied. We showed that, while the Su(Hw) protein remains stably associated with the promoters of these genes at different transcriptional state, the GAF protein level is significantly higher when transcription is activated.

The Antiproliferative Effect of the “Early” Protein E2 of Papillomavirus HPV16 on Testis Tumors of Mice Induced by the Injection of HeLa Cells

Аннотация

The antiproliferative effect of the “early” protein E2 of the high-risk oncogenic human papillomavirus HPV16 on mouse testis tumors, which were induced by the intramuscular injection of HeLa cells, was discovered. The regression of tumors was maximum in the first 2 days after the oral vaccination with HPV16 E2 (500 mg per mouse) and then gradually decreased to the control variant. A typical monolayer of HeLa cells on the cultivation flask bottom, of which only 18% were functionally active, appeared after seeding testis tissue cells on DMEM medium.

Protein Partners of Selenoprotein SELM and the Role of Selenium Compounds in Regulation of Its Expression in Human Cancer Cells

The search for potential partners of human SELM in lysates of two cancer cell lines, HT-1080 (fibrosarcoma) and MCF-7 (breast adenocarcinoma), was carried out. Two cytoplasmic actin isoforms—cytoplasmic actin 1 (cytoskeleton β-actin) and cytoplasmic actin 2 (cytoskeletal γ-actin)—were identified as partners. In addition, the influence of two widely used antitumor selenium compounds (sodium selenite and methylseleninic acid) on the expression of SELM in cancer cells was studied. On the basis of the results obtained by real-time PCR and Western blotting, we concluded that 1 μM and 10 μM sodium selenite did not affect the expression of SELM in fibrosarcoma cells, whereas in breast adenocarcinoma cells 1 μM sodium selenite slightly increased expression and 10 μM sodium selenite significantly (approximately 2 times) decreased it. Methylseleninic acid in both cancer cell lines increased the SELM gene expression; the most pronounced effect was observed when fibrosarcoma cells were treated with 10 µM MSA (the expression of the hSelm gene increased almost 4 times).

New Positive Allosteric Modulator of AMPA Receptors: in vitro and in vivo Studies

Аннотация

A new derivative of 3,7-diazabicyclo[3.3.1]nonane, which showed a high activity as a positive allosteric modulator of AMPA receptors of the CNS, was studied in electrophysiological experiments. At doses of 0.01 mg/kg, this compound significantly improved the memory of experimental animals disturbed by maximal electric shock. The results indicate that this compound is a promising candidate for preclinical trials and clinical studies as a drug for treatment of a number of psychoneurological diseases.

Sigma-1 Receptor Agonist Amitriptyline Inhibits Store-Dependent Ca2+ Entry in Macrophages

Using Fura-2AM microfluorimetry, we have shown for the first time that sigma-1 receptor agonist–tricyclic antidepressant amitriptyline—significantly inhibits store-dependent Ca2+ entry, induced by endoplasmic Ca2+-ATPase inhibitors thapsigargin and cyclopiazonic acid, in rat peritoneal macrophages. The results suggest a possible involvement of sigma-1 receptors in the regulation of store-dependent Ca2+ entry in macrophages.

An Approach to Better Estimation of the Initial Velocity of Enzyme Reactions

Аннотация

A new algorithm for computation of the initial velocity of enzyme reaction at the time zero is proposed. This algorithm makes it possible to reduce the systematic error of measurements to the minimum, to estimate the reaction velocity in test samples regardless of the enzyme activity levels, and to minimize the assay time. The study is illustrated by an example of salivary alpha-amylase and standard reagent kit. The algorithm should not be applied if conjugated enzyme systems are used because there is a long initial lag phase in the kinetic curve.

Effect of Fabomotizole on Brain Gene Expression in MR Rats in the Open Field Test

Selective anxiolytic fabomotizole (Afobazol®) has affinity for the Sigma-1 chaperone receptor site, quinone reductase 2 (NQO2) and MAO-A regulatory sites, and melatonin receptor type 1 (MT1 receptor). The analysis of the effect of fabomotizole on the gene expression profile in the brain of MR (Maudsley Reactive) rats was carried out when modeling emotional stress in the open field test. A change in the expression of 14 genes was found, the results of the functional annotation of which showed that the mechanisms of action of fabomotizole may be associated with the regulation of translation of proteins (Rpl5, Rpl15, Ncl, and Ybx1), synaptic functions (Cplx2, Dlg4, Syngap1, Add1, Rab8b, Klc1, and Chn1), and cellular metabolism (Akr1d1, Bcat1, and Pkm).

High contents of interferon and CD4 and CD8 T lymphocytes were found in blood and spleen cells of mice after their oral vaccination with the “early” protein of HPV16 E2 by using ELISPOT, Western blot hybridization, and enzyme immunoassay technique. It was concluded that the “early” protein HPV16 E2 is involved in the immune system activation during control of pathogenesis and neoplasia.

It was shown for the first time that pentaamino acid derivative of fullerene C60 (potassium salt of fullerenylpenta-N-dihydroxytyrosine) affects three targets of type 2 diabetes mellitus. It competitively inhibits the enzymes aldose reductase and sorbitol dehydrogenase and also has an antiglycation effect on bovine serum albumin. The inhibition constants for these enzymes were calculated.

A New Component in the Mechanism of Regulation of Endogenous Depressive-Like States

Аннотация

It was shown previously that cycloprolylglycine, an endogenous neuropeptide, is a positive AMPA receptor modulator and is able to increase the content of BDNF in neurons. In the present study, using the model of learned helplessness in rats, we showed that cycloprolylglycine at a dose of 1 mg/kg at subchronic intraperitoneal administration reduced the time of animal immobility to passive control values (from 167.6 to 83.6 s) on day 12 of the experiment. This indicates the presence of antidepressant-like activity. The results of the study allow us to consider cycloprolylglycine as a component in the physiological regulation of the depression-like state.

Expression of P-Type ATPases of Marine Green Microalga Dunaliella maritima under Hyperosmotic Salt Shock

Partial sequences of P-type ATPases were cloned from the marine microalgae Dunaliella maritima, two putative H+-ATPases (DmHA1 and DmHA2) and two putative Ca2+-ATPases (DmCA1 and DmCA2). The probable functions of the cloned proteins were suggested on the basis of their primary structure similarity with the proteins whose functions have been already characterized. The transcriptional response of the cloned D. maritima ATPase genes to a sharp increase in the NaCl concentration in the culture medium (from 100 to 500 mM) was investigated by quantitative RT-PCR. Hyperosmotic salt shock led to a significant increase in the DmHA2 expression and to a slight increase in the DmCA2 expression, whereas the expression of the two other ATPases, DmHA1 and DmCA1, was decreased. These data indicate that the DmHA2 ATPase is involved in maintenance of ion homeostasis in D. maritima cells under hyperosmotic salt shock.

Plasminogen, the precursor of plasmin, is a serine protease that plays a fundamental role in the intravascular thrombolysis. For the first time, by using high-resolution mass spectrometry, data on the oxidative modifications of the plasminogen molecule under induced oxidation were obtained. The FTIR data show that, under oxidation on the protein, its secondary structure also undergoes the rearrangements. The high tolerance of plasminogen to oxidation can be due to both the closed conformation and the ability of some Met residues to serve as ROS trap.

Dimeric Disintegrins from the Steppe Viper V. ursinii Venom

Four dimeric disintegrins were isolated from the venom of the steppe viper V. ursinii using liquid chromatography. Disintegrins prevented adhesion of MCF7 cells to fibronectin, which indicates their interaction with integrin receptors of the αVβ1 type. According to mass spectrometry data, the molar masses of disintegrins are about 14 kDa. The method of peptide mapping established the structure of a new heterodimeric disintegrin weighing 13 995.5 Da and shows that it belongs to the class of RGD/KGD-containing disintegrins.

The antioxidant and antiradical properties of the tetra nitrosyl iron complex with thiosulfate ligands (TNIC) were studied in vitro in mouse brain homogenates. It was found for the first time that TNIC is an effective antioxidant. The effect of TNIC on the catalytic activity of mitochondrial enzymes cytochrome c oxidase and monoamine oxidase A was studied. It was shown for the first time that TNIC is an inhibitor of the catalytic activity of cytochrome c oxidase and monoamine oxidase A in animal brain mitochondria in vitro.

Comparative Analysis of the Biosynthesis of Isoprenoid and Aromatic Cytokinins

To compare the biosynthesis pathways of aromatic and isoprenoid cytokinins, a series of nucleoside derivatives of natural cytokinins was synthesized and their cytokinin activity was determined in a test system based on the model plant Arabidopsis thaliana. Cytokinin nucleosides are known to lack the hormonal activity until cleaving the ribose moiety at the position 9. Our experiments have shown that both ribo- and 5'-deoxyribo derivatives of N6-isopentenyladenine were able to turn into active cytokinins in planta exhibiting cytokinin activity. By contrast, 5'-deoxy nucleosides of aromatic cytokinins did not show similar activity. Since 5'-deoxy nucleosides cannot phosphorylate in vivo, the direct pathway of active cytokinin formation by cleavage of nucleotides is blocked here. The detected activity in 5'-deoxy nucleosides of isoprenoid cytokinins and the lack of the activity in 5'-deoxy nucleosides of aromatic cytokinins indicates the difference in the biosynthesis of these compounds.

Effect of Salicylic Acid on the Oxidative and Photosynthetic Processes in Tomato Plants at Invasion with Root-Knot Nematode Meloidogyne incognita (Kofoid Et White, 1919) Chitwood, 1949

A study of the processes of lipid peroxidation and the activity of the peroxidase enzyme, as well as photosynthetic pigments in susceptible tomato plants treated with salicylic acid (SA), during infection with the root-knot nematode Meloidogyneincognita. It was shown that, in the roots of SA-treated plants, the activity of lipid peroxidation is higher compared to the untreated plants, especially in the case of nematode invasion. A significant increase in the activity of lipid peroxidation in SA-treated invasive plants compared with untreated was noted during the transition of larvae to the sedentary stage and the beginning of the formation of feeding areas—giant cells (3–5 days after invasion). This, apparently, contributes to the inhibition of the development of the parasite and the reduction of plant infection and also indicates the involvement of oxidative processes in the mechanism of the induced resistance of plants to root-knot nematodes. In the SA-treated plants, the qualitative and quantitative composition of photosynthetic pigments, disturbed by invasion, was restored and corresponded to the control level.

Efficiency of Penetration of Dopamine and Serotonin Peptide Derivatives through the Artificial Membranes

A mass spectrometric method has been developed for determining the content of dopamine and serotonin derivatives, which allows evaluating the efficiency of their penetration through artificial membranes depending on the structure of their peptide fragment. In this case, the diffusion of dopamine and serotonin derivatives through the membrane occurred as a result of competitive interactions. It was shown which compounds in this mixture more easily penetrate through artificial membranes. It was found that the most promising in terms of overcoming the BBB are Boc–Pro–Srt and Boc–Pro–DOPA.
