Генотоксический стресс как триггер эндотелиальной дисфункции у крыс линии Wistar: результаты молекулярно-генетического исследования

Аннотация
Атеросклероз и ишемическая болезнь сердца являются одними из ведущих причин инвалидизации и смертности среди населения. Показано, что генотоксический стресс потенциально может рассматриваться как новый триггер эндотелиальной дисфункции, лежащей в основе патогенеза данных заболеваний. Цель исследования – оценка молекулярно-генетических маркеров эндотелиальной дисфункции, ассоциированной с генотоксическим стрессом, у нормолипидемических крыс линии Wistar. В исследование были включены самцы крыс линии Wistar, получавшие инъекции алкилирующего мутагена митомицина С (ММС) в дозировке 0.5 мг/кг массы тела (экспериментальная группа) или 0.9%-ного раствора NaCl (контрольная группа) в хвостовую вену три раза в неделю на протяжении месяца. Уровень генотоксического стресса у животных оценивали с помощью микроядерного теста в полихроматофильных эритроцитах (ПХЭ); эндотелиальную дисфункцию идентифицировали путем оценки экспрессии генов Vcam1, Icam1, Sele, Selp, Il6, Ccl2, Cxcl1, Mif, Vwf, Serpine1, Plau, Plat, Klf2, Klf4, Nfe2l2, Nos3, Snai1, Snai2, Twist1, Zeb1, Cdh5 и Cdh2 в смывах эндотелиального монослоя нисходящей аорты. Установлено, что у крыс из экспериментальной группы развивается выраженный генотоксический стресс, о чем свидетельствует более чем трехкратное повышение частоты ПХЭ с микроядрами и снижение доли ПХЭ в общем пуле проанализированных эритроцитов. Анализ профиля генной экспрессии показал, что у крыс, включенных в экспериментальную группу, наблюдается провоспалительная активация эндотелия, сопровождающаяся повышенной экспрессией генов Vcam1, Icam1, Selp, Il6, Ccl2 и Cxcl1, а также нарушение эндотелиальной механотрансдукции, характеризуемое снижением экспрессии генов Klf2 и Klf4. Таким образом, ММС-индуцированный генотоксический стресс у нормолипидемических крыс линии Wistar ассоциирован с нарушением двух ключевых звеньев патогенеза эндотелиальной дисфункции и может рассматриваться в качестве одного из ее триггеров.
