Аннотация
Атопический дерматит ― гетерогенное воспалительное заболевание кожи, на которое влияет множество внутренних (генетических) и внешних факторов. Взаимодействие между генетическими и факторами окружающей среды является сложным динамичным процессом. Эти факторы влияют друг на друга и могут быть взаимосвязанными.
В обзоре рассматриваются данные о филаггрине и некоторых экспосомных факторах, влияющих на него.
Термин «экспосом» включает в себя понятие о влиянии внешних факторов на биологическую среду организма, диапазон которых достаточно разнообразен ― от воздействия химических веществ, психосоциальных и физических факторов до связанных с ними биологических реакций на протяжении всей жизни человека.
В статье обсуждается связь генетической предрасположенности и мутаций филаггрина, на которые потенциальное влияние оказывают дефекты кожного барьера, иммунный дисбаланс и разнообразные триггеры.
Рассматриваются также механизмы действия различных экспосомных факторов на филаггрин при атопическом дерматите. Кроме того, факторы окружающей среды могут влиять на экспрессию различных генов и, соответственно, быть причиной эпигенетических изменений у больных атопическим дерматитом.
Изучение эпигенетических изменений имеет важное значение для уточнения молекулярных основ этого кожного заболевания и улучшения методов его терапии.
