An Inventory of Marine Benthic Macroalgae of Hainan Island, China
Аннотация
In this review, the results of the study of marine benthic algal flora of Hainan Island, China, during the period from 1933 to 2015 are considered. It has been found that the local floristic diversity and taxonomic composition are similar to those of macroalgae from other large islands of the Indo-Pacific, where coral reefs are the main ecosystem of the shallow-water zone. An analysis of historical (decadal) changes in the marine benthic flora of Hainan has shown the following pattern of variations in the species diversity of macroalgae since the middle of the 20th century: a decline in low-productive epilithic algae with voluminous forms of thalli and an increase in highly productive opportunistic species with filamentous, fine filamentous, and membranous forms. These historical changes are thought to be caused by human impacts: inappropriate exploitation of coral reefs and eutrophication of coastal waters due to household sewage from large cities and discharge of mariculture wastes.

The Quantitative Features of the Phytoplankton Community in the Southwestern Kara Sea in the Summer of 2015

This paper presents results from a study of phytoplankton communities that was performed in 2015 at 213 stations in the southwestern part of the Kara Sea. The values of primary production at most stations exceeded the estimates obtained in 1993 by several times. A continued increase in productivity indices and no outbreaks of phytoplankton were consistent with long-term satellite monitoring data. At the same time, grouping of stations showed a highly mosaic structure of the spatial distribution of phytoplankton and a significant range of variation of most of the studied parameters.



Morphological and Genetic Identification of Pseudo-nitzschia H. Peragallo, 1900 (Bacillariophyta) from the Sea of Japan

A total of 15 Pseudo-nitzschia strains isolated from Russian coastal waters of the Sea of Japan were identified using light and electron microscopy and phylogenetic analysis of ribosomal genes. Pseudo-nitzschia species are known to be potential producers of neurotoxiс domoic acid and cause blooms. Phylogenetic analysis of LSU and ITS1-5,8SrDNA-ITS2 sequences revealed that the examined strains belong to P. calliantha, P. hasleana, P. multistriata, P. pungens var. pungens, and P. delicatissima. P. hasleana is a new record for the Sea of Japan. Some morphological distinctions of the strains of P. calliantha and P. pungens var. pungens from the original descriptions of the species were detected. The ITS1-5,8SrDNA-ITS2 topologies showed that the P. delicatissima clade A is formed from three subclades, including Pacific strains, as well as isolates from Scotland and Spain. The high values of genetic distances between subclades (3.7-5%) and the morphological similarity between strains are indicative of cryptic genetic diversity within the P. delicatissima clade A. Different ITS types, including a possibly recombinant type, occur in the same monoclonal culture of P. multistriata, suggesting the hybridization of the local populations in the studied area.



Application of a Method of Multi-Substrate Testing of the State of Marine Surface Waters

The state of waters in Peter the Great Bay, Sea of Japan, and Nha Trang Bay, South China Sea, has been assessed according to the following microbiological parameters at the sites that differ in their degrees of anthropogenic impact: spectra of substrate consumption determined by the method of multi-substrate testing (MST) and the abundance of some indicator bacteria. The results of mathematical processing of the spectra of substrate consumption by microorganisms in the studied waters showed the main characteristics of secure well-functioning water ecosystems and systems under the influence of external factors. The cleanest water sites were characterized by the maximum values of the ecosystem health index and the minimum values of the number of conditionally pathogenic bacteria. Consequently, the data obtained through determination of the spectra of substrate consumption by marine microflora reflect the actual ecological and epidemiological situation in the water area and can be used in monitoring of marine waters.



The Bowhead Whale, Balaena mysticetus Linnaeus, 1758, in the Western Sea of Okhotsk (2009–2016): Distribution Pattern, Behavior, and Threats

The data on sightings of the bowhead whale (Balaena mysticetus Linnaeus, 1758) in the western Sea of Okhotsk collected during eight field seasons, as well as information obtained from interviews with local residents in the study region, are presented. The major areas of concentration of the bowhead whale population in the ice-free period are determined; the whale behavior in bays is described. Data on sea surface temperature, salinity, and depth measurements at the sites of bowhead whale sightings are provided. The possible causes of whale concentrations close to the shoreline and in apical parts of mainland shallow-water bays, as well as the potential threats to the population associated, inter alia, with this pattern of distribution, are discussed.



Dexamethasone Modulation of the Interaction between Two Types of Phagocytes of the Holothurian Eupentacta fraudatrix (Djakonov et Baranova, 1958) (Holothuroidea: Dendrochirotida)
Аннотация
This study examines the interaction between two types of phagocytes (P1 and P2) of the holothurian Eupentacta fraudatrix and its in vitro modulation by dexamethasone. Our results indicate that inhibition of apoptosis in P1 phagocytes by P2 phagocytes was accompanied by increased activities of antioxidant enzymes and reduced synthesis of interleukin-1α-like substances. We hypothesize that P1-phagocyte-related effects occurred in response to a high level of hydrogen peroxide produced by P2 phagocytes. The reduced anti-apoptotic effect of P2-phagocyte supernatant during prolonged incubation (24 h) was accompanied by a decline in defense reactions in P1 phagocytes due to depletion of antioxidant enzymes (catalase, glutathione reductase, and glutathione transferase). Inhibition of apoptotis in P1 phagocytes associated with upregulation of antioxidant enzyme defense in response to P2 phagocytes preincubated with dexamethasone (100 µM) indicates that P2 phagocytes affect P1 phagocytes via a ROS-associated mechanism. Thus, our data provide evidence that P1 and P2 phagocytes exhibit their maximum activity at different stages of the immune response, thus causing inhibition of activity in target cells during prolonged exposure. Dexamethasone enhances these effects.



The Composition of Carotenoids in Tissues of the Ascidian Botryllus schlosseri (Pallas, 1766) from the Black Sea

The composition of carotenoids was investigated in tissues of the colonial sea squirt Botryllus schlosseri (Pallas, 1766), which inhabits the Crimean coast near the city of Sevastopol. The total carotenoid amount of B. schlosseri was found to be 8.7 ± 4.6 mg/100 g of wet weight. Eleven carotenoids, that is, ß-carotene, 7,8-didehydroastaxanthin, 7,8,7',8',-tetradehydroastaxanthin, pectenolone, 4-ketoalloxanthin, diatoxanthin, diadinoxanthin, fucoxanthin, astaxanthin, alloxanthin, and halocynthiaxanthin, were identified in B. schlosseri with the use of the chromatographic and mass-spectrophotometric methods (UV/Vis, ESITOF/MS, and partially 1H-NMR). Among these, halocynthiaxanthin (20.8% of the total content of carotenoids), alloxanthin (15.2%), and astaxanthin (12.1%) were found to be the major carotenoids. A comparative analysis of the composition of carotenoids in tissues of B. schlosseri was carried out for different regions of the World Ocean.



A Comparative Study of Free Amino Acids of the Brown Alga Fucus vesiculosus Linnaeus, 1753 from the Intertidal Zone of the Murman Shore, Barents Sea

The free amino-acid contents of the apical and middle parts of the thallus, as well as the receptacles of fertile and juvenile specimens, of the brown alga Fucus vesiculosus collected from seven different biotopes of the Murman shore, Barents Sea have been studied for the first time. The free amino-acid composition of the studied algae specimens was dominated by alanine, along with aspartic and glutamic acid. The distribution of free amino acids in the thallus of Fucus is uneven and depends on the habitat. The free aminoacid contents in the spring vary considerably between different parts of the thallus; however, certain regular patterns of their distribution are observed. A correlation has been found between the level of some individual amino acids in different parts of the thallus of fertile and juvenile algae and the salinity of the seawater.



The Influence of Phytoplankton Primary Production on the Cycle of Biogenic Elements in the Coastal Waters off Sevastopol, Black Sea
Аннотация
The phytoplankton primary production and the biogenic elements content of the coastal waters off Sevastopol have been studied as a result of 2-year monitoring. It has been found that mineral phosphorus is a key factor of chemical limitation of the production processes. During primary production of organic matter, the habitat conditions for phytoplankton in the photic layer are in accordance with the Le Chatelier–Braun principle of negative feedback.



High-level Recombinant Expression and Refolding of Lysozyme from the Marine Shrimp Marsupenaeus japonicus

Shrimp lysozyme is as an antibacterial enzyme that participates in the innate defense against the invasion of bacterial pathogens. In this study, the lysozyme gene from hemocytes of the shrimp Marsupenaeus japonicus was isolated and characterized. The M. japonicus lysozyme (MjLys) encodes a polypeptide of 158 amino acids (aa) that includes an 18 aa signal peptide. The gene fragment encoding the mature MjLys protein was subcloned into the expression vector pET-32a(+) and transformed into E. coli BL21(DE3)pLysS, and the protein was strongly expressed in insoluble inclusion bodies. Following extraction using urea, the denatured recombinant protein was refolded by on-column Ni2+ affinity chromatography or dialysis with a gradient of decreasing urea concentration. Approximately 50% of the recombinant MjLys was successfully refolded into monomeric protein using urea gradient dialysis, while 30% was salvaged using on-column refolding. Purified MjLys exhibited significant antibacterial activity against Gram-positive bacteria Micrococcus lysodeikticus and Staphylococcus aureus. This efficient over-expression and refolding method can provide the large quantities of biologically active protein required for further biochemical and structural studies and potential biotechnological applications.



On a Poorly Known Rookery of Green Turtles (Chelonia mydas) Nesting at the Chabahar Beach, Northeastern Gulf of Oman

There is a poorly known rookery of green turtles (Chelonia mydas) nesting in sandy beaches of Chabahar town, northeastern Gulf of Oman, Iran.This study has been carried out to evaluate nesting activity of this small rookery in 2014 nesting season (June to October). In this study, total clutches were collected and transferred to an artificial hatchery. The peak of nesting occurred from the third parts of August to the end of September. Mean CCL was 106.3 ± 6 cm and mean CCW was 94.5 ± 5 cm. Females laid on average of 99.42 ± 47.8 eggs per clutch. The mean inter-nesting interval was 18.5 days. The observed clutch frequency was 3.4. Mean hatching success was 36.63 ± 4.1%. The incubation period was 61.07 ± 5.4 days. Nest status evaluation represented that major causes for the failure was unhatched egg with no obvious embryo followed by unhatched eggs with obvious but undeveloped embryos > unhatched eggs with developed embryos > hatched eggs but dead embryos. The results achieved in this study are a valuable contribution to cognition of the reproductive ecology of the green turtle population globally and regionally.
