Обсуждаются существующие представления о молекулярных механизмах патологической гипервозбудимости и синхронизации нейронных сетей при эпилептогенезе, включая калиевую, ГАМК-, мембранную (клеточную) и синаптическую (сетевую) модели. В фокусе таких моделей – нарушение баланса между возбуждением и торможением с вовлечением многочисленных положительных и отрицательных обратных связей в нейронных сетях. В работе рассматриваются современные представления о: 1) надежности динамических систем с большим числом отрицательных обратных связей и 2) вырожденности, т.е. способности разнородных элементов (каналов, токов) замещать друг друга, как основе устойчивого функционирования гипервозбудимых сетей при каналопатиях и гиперэкспрессии различных каналов. В работе предлагается возможный механизм спонтанного возникновения судорожной активности и накопления калия в межклеточном пространстве, основанный на активации калием и кальцием группы катионных каналов (HCN, Kir2.x, hERG, Nav1.х и ВКСа), обеспечивающий надежность и высокую чувствительность эпилептиформной активности к внешним и внутренним факторам за счет вырожденности и формирования группы петель положительных обратных связей.
