МикроРНК как перспективные биомаркеры при раке

Аннотация
К 2004 г. в результате международного проекта по секвенированию генома человека было проанализировано около 20 000 кодирующих белок генов, что соответствует 2 % общей геномной последовательности. Подавляющее большинство транскриптов генов фактически характеризуются как некодирующие РНК (нкРНК) и представляют собой кластеры РНК, которые не кодируют функциональные белки. Они могут быть небольшими, длиной примерно 20 нуклеотидов, и известны как микроРНК (miRNAs), или транскриптами длиной более 200 нуклеотидов, определяемыми как длинные нкРНК (lncRNAs). miRNA — это короткие нкРНК, участвующие в посттранскрипционной регуляции экспрессии генов. Обнаруженные более 15 лет назад эти молекулы признаны в качестве одних из основных регуляторных молекул в геноме человека. Они играют важную роль во всех биологических процессах и модулируют экспрессию эукариотических генов. Ориентируясь на транскрипты, кодирующие белки, miRNA влияют на клеточный транскриптом, тем самым помогая определить судьбу клетки. Была замечена аберрантная экспрессия miRNA у больных раком. Оказалось, что тканевые концентрации специфических miRNA связаны с опухолевой инвазивностью, метастатическим потенциалом и другими клиническими характеристиками для многих типов рака. По сравнению с традиционными биомаркерами, такими как белки, miRNA имеют ряд преимуществ, которые позволяют использовать их в качестве новых потенциальных биомаркеров при раке. В этом обзоре рассмотрен биогенез, функции, технологии, применяемые для обнаружения miRNA, и ассоциации микроРНК с раком человека, в частности колоректальным раком.
