Аннотация
В обзоре литературы затронуты вопросы применения метаболомических исследований в медицине. Основная идея метаболомики заключается в обнаружении специфических биомаркёров в биологическом образце для диагностики ряда заболеваний. В качестве биомаркёров рассматриваются летучие органические вещества - метаболиты, выделенные из различных биологических тканей и жидкостей (крови, мочи, мокроты, выдыхаемого воздуха). Отражены основные методы разделения и идентификации летучих органических веществ образца (газовая хроматография, масс-спектрометрия, спектроскопия ядерного магнитного резонанса), применяемые в метаболомике. Даны сравнительные характеристики масс-спектрометрии и спектроскопии ядерного магнитного резонанса как основных методов детектирования летучих метаболитов. Описан метод твердофазной микроэкстракции, используемый в качестве предварительной подготовки образца. Представлены результаты лабораторных исследований по поиску биомаркёров рака, хронических инфекций, наследственных заболеваний. Показаны качественные характеристики метаболома биологического образца пациента с той или иной патологией. Кроме того, уделено внимание применению возможностей метаболомики в экспериментальной медицине. Описаны результаты исследования изменений летучего метаболома клеточной культуры in vitro в зависимости от добавок в питательные среды; летучих продуктов распада β-каротина как проканцерогенных веществ; летучих органических веществ, выделяемых при распаде позвоночных животных. Дополнительно описан метод полной двухмерной газовой хроматографии, испытанный с целью повышения чувствительности и специфичности метаболомических исследований. Представленный подход может помочь в решении вопросов ранней диагностики многих заболеваний.
