АНАЛИЗ НЕОПРЕДЕЛЕННОСТЕЙ РАСЧЕТОВ ЭФФЕКТИВНОСТИ РАДИАЦИОННЫХ ПОВРЕЖДЕНИЙ ДНК

Аннотация
Компьютерные вычисления эффективности образования радиационно-индуцированных повреждений ДНК достаточно широко востребованы в радиобиологии и биомедицине. Методы расчета основаны на молекулярных представлениях об образовании разрывов, структуре ДНК и информации о распределении поглощенной энергии излучения в ядре клетки. Поскольку расчетные данные используют разные предпосылки, постулаты и алгоритмы, сопоставление точности расчетов по разным схемам часто затруднено, как и определение параметров моделей разрывов с учетом разброса экспериментальных данных. В данной работе проведен сравнительный анализ предсказаний двух базовых моделей структуры ДНК, молекулярной и субобъемной, использующих различные предположения о механизме образования одно- и двунитевых разрывов ДНК, на примере облучения фрагментов ДНК протонами и α-частицами. Расчеты эффективности радиационных повреждений ДНК, учитывающие вклады прямого и квазипрямого действия, молекулярную структуру ДНК и стохастическую структуру трека заряженных частиц, показывают зависимость частот повреждений ДНК от параметров моделей. Найдены области параметров, где предсказания моделей согласуются или отличаются друг от друга.
