На переживающих срезах гиппокампа крыс методом парной стимуляции исследовали изменение свойств пресинаптической кратковременной пластичности на фоне блокады деполимеризации и активации полимеризации актина. Тестирование синаптической передачи СА3-СА1 в режиме парной стимуляции с межстимульным интервалом 70 мс производилось до и на фоне джасплакинолида, ингибитора деполимеризации, применяемого также для индукции полимеризации актина. Увеличение амплитуды ответа при аппликации джасплакинолида сопровождалось снижением фасилитации при парной стимуляции (PPF), что свидетельствует о пресинаптических механизмах потенциации. Эффект усиливался пропорционально величине исходной PPF. Учитывая известные механизмы, лежащие в основе PPF, можно предположить, что при низкой вероятности выброса медиатора дополнительная полимеризация актиновых филаментов способствует её увеличению. Иначе изменялись менее типичные для поля СА1 реакции на парную стимуляцию (PPF около 1 или даже парная депрессия), и джасплакинолид увеличивал амплитуду второго, но не первого в паре ответа, так что соотношение амплитуд второго и первого ответа даже увеличивалось в среднем с 0,8 до 1. Предполагается, что, помимо основного эффекта, на фоне джасплакинолида снижается влияние факторов, способствующих парной депрессии. Так или иначе, полимеризация актина приводит к потенциации синаптических реакций, хотя паттерн активации может различаться в зависимости от исходных свойств пресинаптической пластичности. В заключение: комплексный эффект джасплакинолида, помимо увеличения вероятности выброса медиатора, включает и другие зависимые от полимеризации актина механизмы, в том числе лежащие в основе кратковременной синаптической депрессии при парной стимуляции.
