Эпителиально-мезенхимальный переход клеток рака молочной железы при активации транскрипционного фактора SNAIL1

Аннотация
Программа эпителиально-мезенхимального перехода (ЭМП) используется опухолевыми клетками при активации инвазионно-метастатического каскада. С помощью конфокальной и видеомикроскопии нами изучены изменения цитоскелетных структур клеток линии рака молочной железы MCF-7 в ходе ЭМП при активации транскрипционного фактора Snail1. В работе была использована линия клеток MCF-7, стабильно экспрессирующих tetOff-регулируемую конструкцию SNAI1 (MCF-7-SNAI1). При отмывке от тетрациклина и активации Snail1 клетки MCF-7-SNAI1 вступали в ЭМП и приобретали миграционный фенотип, сохраняя при этом экспрессию Е-кадхерина. Мы выделили пять вариантов мезенхимального фенотипа, различающихся по морфологии клеток и скорости миграции. Мигрирующие клетки обладали выраженной пластичностью и были способны быстро изменять как фенотип, так и скорость движения. Как показала иммунофлуоресцентная микроскопия, в основе изменений фенотипа клеток MCF-7-SNAI1 при ЭМП лежат Arp2/3-зависимая полимеризация актиновой сети в ламеллиподиях на ведущем крае, активация контрактильности миозина в зоне за ядром, перераспределение белков межклеточной адгезии из зон межклеточного взаимодействия на ведущий край и реорганизация сети промежуточных кератиновых филаментов.
