Физико-химические особенности связывания тромбина с мембраной тромбоцита

Тромбин – ключевой фермент системы свертывания крови, который активно изучается с начала прошлого века. Образование тромбина из протромбина в области повреждения сосуда приводит не только к формированию фибрина – важного структурного компонента гемостатического сгустка, но и к активации тромбоцитов, эндотелия и клеток имMунной системы. Считается, что связывание тромбина с поверхностью тромбоцитов играет критическую роль в процессе их активации, а также может обеспечивать поддержание высокой концентрации тромбина внутри тромба вследствие концентрирования протеазы на поверхности тромбоцитов. На данный момент все основные рецепторы тромбина на тромбоцитах тщательно охарактеризованы: благодаря различным экспериментальным методам установлены физико-химические параметры соответствующих межмолекулярных взаимодействий. Так как взаимодействие тромбина с тромбоцитами приводит к их активации, включающей изменения количества рецепторов в результате секреции гранул, правильная интерпретация наблюдаемых особенностей кинетических кривых связывания сильно усложняется. Известно, что некоторые рецепторы в результате активации тромбоцита способны перераспределяться на мембране и образовывать димеры и кластеры, что делает кинетику связывания тромбина с тромбоцитами чрезвычайно сложным процессом, зависящим от множества факторов, таких как концентрации активаторов, состояние тромбоцитов и других локальных параметров системы. Данный обзор ставит своей целью описать современные представления о взаимодействии тромбина с мембраной тромбоцита и обозначить важные нерешенные вопросы в данной области исследований. В обзоре представлена не только информация о структурных и кинетических особенностях связывания тромбина с отдельными мембранными белками тромбоцита, но и анализ связи между соответствующими параметрами взаимодействия и ранее полученными данными о кинетике связывании протеазы с поверхностью тромбоцита.
