Анализ медленной инактивации каналов Nav1.5 в развитии наследственной патологии сердца

Аннотация
Потенциал-зависимые натриевые каналы сердца Nav1.5 ответственны за инициацию и распространение потенциала действия в кардиомиоцитах. Дисфункция Nav1.5 может быть обусловлена как патогенными вариантами в самом гене SCN5A, кодирующем Nav1.5, так и генетическими вариантами в генах других белков, регулирующих его активность и транспорт. Смена различных фаз потенциала действия определяется строгой временной организацией активации и инактивации различных ионных каналов. На переходы между функциональными состояниями канала, в том числе на переход в состояние медленной инактивации, могут влиять разнообразные факторы и взаимодействующие с каналом белки. Хотя процесс медленной инактивации канала известен несколько десятилетий, его роль в механизме развития наследственной патологии сердца остается неясной. В нашей работе мы с помощью метода локальной фиксации потенциала (patch clamp) в отведении от целой клетки (whole-cell) исследовали изменения процесса медленной инактивации Nav1.5 под влиянием различных мутаций в структурных генах (DSP-H1684R, LMNA-R249Q, FLNC-R1267Q, FLNC-V2264M), ассоциированных с генетически обусловленной патологией миокарда, приводящей к дисфункции кардиомиоцитов. Мы использовали модель кардиомиоцитов, дифференцированных из индуцированных плюрипотентных стволовых клеток (КМ-иПСК) и продемонстрировали усиление медленной инактивации каналов на модели КМ-иПСК, полученных от пациентов с фенотипом кардиомиопатии, совмещенной с желудочковыми аритмиями. Таким образом, представленная работа вносит вклад в понимание роли процесса медленной инактивации Nav1.5 в механизме развития патологии сердца.
