Получение и первичная оценка фенотипа тополя берлинского, трансформированного геном AtGA20ox1

Увеличение скорости набора биомассы является одним из важнейших направлений в селекции древесных растений. Тем не менее, применение классических подходов селекции к древесным растениям существенно ограничено из-за длительных циклов размножения многих видов. Развитие технологий генетической инженерии и редактирования генома позволило проводить улучшение признаков деревьев за относительно короткое время. Объектом для генетических манипуляций, целью которых является ускорение роста растений, часто являются гены биосинтеза фитогормонов. Гиббереллин-20-оксидаза является ключевым ферментом, определяющим активное производство гиббереллинов в растениях и, следовательно, предпочтительной мишенью для генетических манипуляций, направленных на увеличение скорости роста. В данной работе представлено оригинальное исследование по получению тополя берлинского Populus × berolinensis K. Koch, трансформированного геном, кодирующим гиббереллин-20-оксидазу из Arabidopsis thaliana (L.) Heynh. (AtGA20ox1), и первичной оценке фенотипических эффектов проведенной трансформации. Основными фенотипическими проявлениями трансформации явились выраженное удлинение стебля за счет увеличения размера междоузлий, незначительное его истончение, а также удлинение и заужение листьев. Наши результаты показали, что гиперэкспрессия гена AtGA20ox1 у тополя берлинского приводит к ускорению его роста как минимум в три раза в условиях in vitro по сравнению с контрольными значениями. Негативные эффекты трансформации, выражающиеся в слабом укоренении или высокой частоте апикальных некрозов и наблюдавшиеся у некоторых трансгенных линий на начальных этапах отбора, не проявляются у трех финально отобранных линий.
