РОТЕНОН, РОДАМИН 123 И ЯНУС ЗЕЛЕНЫЙ ИНДУЦИРУЮТ ПОВРЕЖДЕНИЕ ЯДЕРНОЙ ДНК В КЛЕТКАХ АСЦИТНЫХ ОПУХОЛЕЙ МЫШЕЙ, РОТЕНОН И РОДАМИН В Х-ОБЛУЧЕННЫХ КЛЕТКАХ СПОБСТВУЮТ СОХРАНЕНИЮ ЦЕЛОСТНОСТИ ГЕНОМА

Митохондриальные ингибиторы ротенон, родамин 123 и краситель янус зеленый В изучаются в настоящее время с целью разработки фармакологических средств, вызывающих митохондриальную дисфункцию и апоптоз. Поскольку нарушение работы митохондрий связано с продукцией активных форм кислорода, то представляется актуальным сопоставить индуцированные этими веществами повреждения ДНК клеток асцитной карциномы Эрлиха и мышиного лимфолейкоза Р388 с таковыми после воздействия известного индуктора активных форм кислорода – ионизирующего (рентгеновского) излучения. Уровень ДНК-повреждений оценивали щелочным вариантом метода ДНК-комет. Индуцированный ротеноном уровень повреждений ДНК был сравним с таковым при 4 Гр в обоих типах клеток. Пострадиационная инкубация уменьшала уровень ДНК-повреждений, что свидетельствовало о репарации ДНК. Обработка клеток асцитной карциномы Эрлиха родамином 123 с последующей отмывкой от него не вызывала этого увеличения, однако облучение в дозе 4 Гр в присутствии родамина 123 индуцировало увеличение уровня повреждений ДНК, которое существенно уменьшалось после часовой инкубации. Можно полагать, что предварительная обработка клеток ротеноном или родамином 123, нарушая работу митохондрий, способствовала сохранению целостности ядерной ДНК в облученных клетках. Воздействие януса зеленого В вызывало увеличение повреждений ДНК и гибель клеток. Исходя из щелочной версии метода ДНК-комет, можно считать индуцированные этими соединениями повреждения одно-, двунитевыми разрывами и щелочелабильными (апуриновыми/апиримидиновыми) сайтами в ДНК.
