Перспективы изучения нейрохимии и фармакологии игрового расстройства поведения

Аннотация
В настоящее время не существует общепринятых подходов к терапии игровой зависимости, как нет и ни одного зарегистрированного по данному показанию лекарственного средства. Для лечения используют психотерапевтические подходы и фармакотерапию с использованием антидепрессантов, антагонистов опиоидных рецепторов, нормотимиков, антагонистов N-метил-D-аспарагиновой кислоты и антипсихотиков. В обзоре проанализированы перспективы изучения нейрохимии и фармакологии игрового расстройства поведения. Наибольший вклад в функционирование состояния азартной игры вносят дофаминергическая и серотонинергическая системы. При этом особый интерес вызывают эффекты активации D3-рецепторов дофамина, серотониновых рецепторов 5-HT1a, 5-HT2a и 5-HT1b. Для лечения игровой зависимости перспективным является изучение эффектов антагонистов глутаматных рецепторов α-амино-3-гидрокси-5-метил-4-изоксазолпропионовая кислота (AMPAR). Недостаточно изученными остаются системы нейропептидов, в частности, эндогенных опиоидов. Наиболее перспективным представляется также изучение нейропептидов грелина, орексина, пептидов группы нейротрофинов, кортиколиберина CRF, окситоцина, нейропептида Y, глюкагоноподобного пептида GLP-1 и кисспептина. Направленный поиск пептидных мишеней, влияющих на медиаторные системы головного мозга, могут выявить важные звенья для коррекции пристрастия к азартным играм (патологический гэмблинг) и расстройства вследствие пристрастия к компьютерным играм (патологический гейминг).
