Фактор дифференцировки роста GDF11 как потенциальная мишень для лечения заболеваний, ассоциированных со старением

Аннотация
В статье представлен обзор литературных источников, посвященных физиологической роли и функциям некоторых белков суперсемейства TGFβ, а именно GDF11 и GDF8, а также их места в патогенезе ряда заболеваний, риск которых увеличивается с возрастом. Описано возможное терапевтическое использование указанных протеинов. Показано, что роль GDF11 в патогенезе описанных заболеваний неоднозначна. GDF11 является ранее нераспознанным регулятором ремоделирования кости, предотвращает гипертрофию миокарда, а также улучшает состояние животных с экспериментальным сахарным диабетом или нейродегенерацией. Антипролиферативное действие GDF11 также наблюдается при многих онкологических заболеваниях. Однако GDF11 может оказывать негативное влияние на метаболизм мышечной и костной ткани, что может являться ограничением для его применения при некоторых состояниях. Ввиду различий в экспрессии и функции GDF11 в сердечной, нервной, мышечной и других тканях, его разнонаправленном действии и узком терапевтическом диапазоне рекомбинантного GDF11, необходимы дальнейшие исследования для выявления оптимального спектра показаний и ограничений, дозирования и способов снижения побочного действия.
