Ферментативный золь–гель переход в молоке

Целью работы является улучшение понимания механизма ферментативного гелеобразования в молоке за счет установления статистически достоверных изменений вязкости молока и соответствующих электронно-микроскопических исследований процесса структурообразования на ферментативной стадии коагуляции при использовании различных ферментных препаратов. Использование неразрушающего (Hot-Wire) метода контроля вязкости позволило подтвердить снижение вязкости в середине этой стадии и выявить ранее не описанный пик изменения вязкости в конце ферментативной стадии. Параллельно проведенные исследования с использованием трансмиссионной электронной микроскопии и различных методов подготовки препаратов позволили установить, что в конце ферментативной стадии гелеобразования в молоке запускается иерархический процесс преобразований белковой составляющей молока. Спусковым механизмом этого процесса является кооперативный конформационный переход в кластерах мицелл казеина, который вызывает цепочку все более энергоемких реакций, результатом которых является преобразование слабосвязанных кластеров мицелл в более плотные агрегаты. Окончательная структура молочного геля формируется преимущественно из образовавшихся ранее отдельных агрегатов мицелл. Отмечено, что изменений микроструктуры молочного геля на ферментативной стадии гелеобразования при использовании молокосвертывающих ферментов (МФ) животного, растительного и микробного (ГМО) происхождения обнаружено не было. Исследования молекулярно-массового распределения растворимых белковых веществ образцов, произведенных с разными типами МФ, показали, что фермент, выделенный от грибов Rhizomucor miehei, обладает большим протеолитическим действием на белки молока в сравнении с другими вариантами МФ.
