Реакция штамма Bifidobacterium longum subsp. infantis ATCC 15697 на окислительный стресс

Бифидобактерии являются преобладающей популяцией в микробиоте кишечника человека. Несмотря на растущее число данных о полезных свойствах бифидобактерий для здоровья человека, знаний об их антиоксидантном потенциале по-прежнему недостаточно. Роль антиоксидантного потенциала бифидобактерий в поддержании гомеостаза микробиоты кишечника организма хозяина в целом – важнейшая задача, требующая решений. Впервые представлены результаты геномного, транскриптомного и протеомного анализов штамма Bifidobacterium longum subsp. infantis ATCC 15697 после действия окислительного стресса. Растущую культуру штамма подвергали воздействию перекиси водорода в течение двух часов и кислорода в течение двух и четырех часов. Предварительный анализ генома штамма показал наличие 17 генов, кодирующих белки с известной антиоксидантной функцией, как и в других геномах B. longum subsp. infantis, доступных в международной базе данных NCBI. Полнотранскриптомный анализ выявил увеличение уровней транскриптов более чем в 2 раза для шести генов с известной антиоксидантной функцией. Данные количественного протеомного анализа показали повышение уровней белков более чем в 2 раза для пяти ферментов с известной антиоксидантной функцией. Более 28 других белков с повышенными уровнями более чем в 2 раза были идентифицированы в клетках растущей культуры штамма в ответ на продолжительное действие кислорода. Эти белки могут быть вовлечены в процессы ответа клетки на стресс, метаболизм аминокислот, нуклеотидов и в транспортные процессы. В клетках после действия стресса выявлен высокий уровень у шести белков с неизвестными функциями, которые могут играть значимую роль в антиоксидантной защите анаэробных бифидобактерий. Полученные данные предполагается применить при отборе штаммов B. longum subsp. infantis и создании на их основе фармабиотиков, способных корректировать состав микробиоты.
