АННОТАЦИЯ НОВОГО НИЗКОПОРОГОВОГО ПОТЕНЦИАЛЗАВИСИМОГО КАЛЬЦИЕВОГО КАНАЛА ТРИХОПЛАКСА Trichoplax adhaerens (ТИП Placozoa)

Аннотация
Изучение потенциалзависимых кальциевых каналов проливает свет на формирование систем, ответственных за сопряжение сенсоров и актуаторов в живой клетке. На основании данных по потенциалчувствительному кальциевому каналу TCav3 (2063 аминокислотных остатка) из клеток Trichoplax adhaerens идентифицированы гомолог длиной 2090 аминокислотных остатков в скаффолде Trichoplax sp. Н2 и неполный полипептид длиной 1510 аминокислотных остатков в скаффолде Trichoplax adhaerens. Последний гипотетический белок аннотирован как Cav3-канал. Для всех трех белков найден EEDD селективный фильтр и реконструирована коровая структура кальциевого канала, состоящая из 24 трансмембранных α-спиралей. Тем не менее исследуемые белки отличаются цитоплазматическими доменами, что свидетельствует о различной специализации Cav3-каналов при проведении сигнала внутрь клетки. Так, часть AID-мотива (alphainteracting domain) и прилежащего сенсора потенциала из аннотированного канала имеют гомологии в 25 видах костистых рыб, а соответствующая область из обоих других каналов в 41 виде костистых рыб и в 4 видах змей. Существенно, что ниже от AID-мотива костистых рыб, как и у трихоплакса, обнаружена высококонсервативная петля IIS1-S2 с последовательностью IEHHNQP, в то время как у змей найдена гомологичная последовательность IEHHEQP, отличающаяся отрицательным остатком глутаминовой кислоты, который также присутствует в соответствующих белках крысы и человека. На основании анализа первичных транскриптов и зрелых полипептидов предполагается модульный механизм эволюции Cav3-каналов путем вставок и объединения белковых доменов, выполняющих разнообразные регуляторные функции.
